Tryptophanyl transfer RNA synthetase from lymphocytes of human chronic lymphocytic leukemia.

چکیده
Tryptophanyl-tRNA synthetase has been purified from a lymphocyte-rich fraction of blood cells of patients with chronic lymphocytic leukemia. Normal lymphocytes, other blood cells, and skin also contain the enzyme. After a 240-fold purification by column chromatography, a further purification was obtained by isoelectric focusing, which revealed an isoelectric point of 5.2 to 5.3. The enzyme is stabilized by 2-mercap toethanol and inactivated by 5, 5 '-dithiobis(2-nitrobenzoic acid). The enzyme is stable in, but inhibited by, phosphate buffer. Yeast tRNATrp is a good substrate, with a Km of 1.1 X 10~7 M. ATP and tryptophan have Km values of 1.6 X 10~4 and 2.9 X 1(T6 M, respectively.

Analysis of HLA-G Gene Expression in B-Lymphocytes from Chronic Lymphocytic Leukemia Patients
The human leukocyte antigen G (HLA-G) molecule exhibits limited tissue distribution, low polymorphism and alternative splicings that generate seven HLA-G isoforms. HLA-G exerts multiple immunoregulatory functions. Recent studies indicate an ectopic up-regulation in tumor cells that may favor their escape from anti-tumor immune responses. This study it is an effort to clarify the presence of HLA...
متن کاملComparison Cytotoxic Effects of Mangifera Indica L. and Juglans Regia Aqueous Extract on Human Chronic Lymphocytic Leukemia
Natural products isolated from plant sources are well known for their pharmacological potential in diversity of disease treatments such as inflammatory or cancer conditions. Mango (Mangifera indica L.) and Juglans regia are thought to be rich of functional phytochemicals. To elucidate the anticancer activity of Juglans regia (JR) and Mangifera indica L (MI) aqueous extract were investigated on ...

متن کاملBiochemical and genetic characterization of a temperature-sensitive, tryptophanyl-transfer ribonucleic acid synthetase mutant of Bacillus subtilis.
A temperature-sensitive, 5-fluorotryptophan (5FT)-resistant mutant of Bacillus subtilis was isolated which forms an altered tryptophanyl transfer ribonucleic acid synthetase [l-tryptophan: sRNA ligase (AMP), EC 6.1.1.2]. The mutant grows well at 30 C but not at 42 C. At the latter temperature, protein and ribonucleic acid (RNA) synthesis are abolished while deoxyribonucleic acid (DNA) synthesis...
